Liver – fatty liver (mouse and human)
Movie 1: Vasculature system in mouse liver 1
Tissue/pathology:
12w male C57BL/6Crl control mouse liver
Antibodies:
Inverted 488 autofluorescence
Movie description:
Vasculature is visualised based on inverted autofluorescence. The portal triad can be clearly identified and discriminated, since three tubes are seen in the triad (one thick: portal vein; and two thin ones: hepatic artery and bile duct) while we see only one thick tube (the vein itself) in case of the hepatic vein.

Tissue/pathology:
12w male C57BL/6Crl control mouse liver
Antibodies:
Inverted 488 autofluorescence
Movie description:
Same scan as in MOVIE 1 but surfaces are created in order to better discriminate between the portal triad (gray) and the hepatic vein (blue).

Tissue/pathology:
12w male C57BL/6Crl control mouse liver
Antibodies:
Cytokeratin 7
Movie description:

Tissue/pathology:
Mouse liver, model for steatohepatitis (SNAP-25b–deficient mice back-crossed on C57BL/6Crl), 12w old male, after 7 week Western diet.
Antibodies:
Cytokeratin 7
Movie description:
Bile duct proliferation (ductular reaction) is well visible. MOVIE 3 is the corresponding control.

Tissue/pathology:
Human healthy liver, 1mm3 cube
Antibodies:
Inverted 488 autofluorescence
Movie description:
Only the contour of vessels and sinusoids are seen.

Tissue/pathology:
Human steatohepatitic liver, 1mm3 cube
Antibodies:
Inverted 488 autofluorescence
Movie description:
Lots of large fat droplets are seen in hepatocytes. I coloured this and the previous videos to yellowish, to imitate the colour of fat. MOVIE 5 is the corresponding control.

Tissue/pathology:
Healthy human liver
Antibodies:
Cytokeratin 7
Movie description:
Demonstration of bile ducts in healthy condition.

Tissue/pathology:
Steatohepatitic human liver
Antibodies:
Cytokeratin 7
Movie description:
The demonstration of bile duct proliferation in steatohepatitic condition (ductular reaction, just like earlier in the respective animal model, MOVIE 4) are well visible. MOVIE 7 is the corresponding control.

Tissue/pathology:
Healthy human liver
Antibodies:
Tyrosine hydroxylase
Movie description:
Note the fine intraparenchymal innervation as well as thicker fibers in the intra-lobular septi and the major fibers around the portal triads. Control (healthy) condition, no nerve pathology is noticed. Also see: liver innervation project.

Tissue/pathology:
Steatohepatitic human liver
Antibodies:
Tyrosine hydroxylase
Movie description:
Note that the fine intra-parenchimal innervation is severely degenerating in steatohepatitis, while thicker fibers in the intra-lobular septi as well as major fibers around the portal triads exhibit swollen axonal pathology suggesting impaired axoplasmatic transport. MOVIE 9 is the corresponding control. Also see: liver innervation project.

Tissue/pathology:
12w male C57BL/6Crl control mouse liver
Antibodies:
Cytokeratin 7 (red); smooth muscle actin (SMA, green)
Movie description:
Smooth muscle actin (green) depicts the major blood vessels. Thin vessel with strong staining: hepatic artery. Thick vessel with moderate intensity staining: portal vein. Thick vessels with faint staining: hepatic vein. Cytokeratin 7 depicts the complex ductular system of intrahepatic bile ducts. 2:30: a ’tour’ inside a portal vein. 3:35: travelling above a portal triad.
